What it is / mechanism

Gonadorelin is a synthetic decapeptide with the identical amino acid sequence to endogenous gonadotropin-releasing hormone (GnRH). It is reported to act as an agonist at GnRH receptors on the anterior pituitary gonadotroph cells, prompting release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). In turn, LH and FSH are reported to signal the gonads to produce testosterone and support spermatogenesis in males, and to regulate follicular development and ovulation in females. A defining feature described in the literature is frequency dependence: pulsatile delivery that mimics natural hypothalamic secretion (roughly every 60-120 minutes) is reported to sustain gonadotropin output, whereas continuous exposure paradoxically downregulates and desensitizes the receptor, suppressing LH and FSH. Its very short half-life is central to this behavior, causing each dose to act as a brief pulse rather than a sustained signal.

Researched effects

In research and clinical reports, gonadorelin has been associated with acute increases in circulating LH and FSH following a pulse, and pulsatile administration has been used to attempt restoration of hypothalamic-pituitary-gonadal (HPG) axis signaling in hypogonadotropic hypogonadism. In community TRT-adjunct discussions it is reported to be used in an attempt to maintain testicular signaling during exogenous testosterone use, as an alternative to hCG. Reconstitution — bac-water math

Reconstitution is concentration math only, not a personal dose. Gonadorelin is dosed in micrograms (mcg), so the useful figure is mcg per milliliter. With a vial of V mg and W mL of bacteriostatic water, concentration = (V x 1000) / W mcg/mL. On a U-100 insulin syringe, 100 units = 1 mL, so units for a dose D (mcg) = round(D / concentration x 100). Worked example: a 2 mg vial reconstituted with 2 mL of water gives 1000 mcg/mL; a 100 mcg dose is 100 / 1000 x 100 = 10 units on a U-100 syringe.

Reading a Gonadorelin certificate of analysis

A certificate of analysis (COA) is a laboratory’s report on one sample of one batch. The single most useful thing to know about it is that purity and identity are two separate results that fail in different ways. A high purity figure says the sample was mostly one substance; it does not say that substance was Gonadorelin. Identity — normally a mass-spectrometry result matching the expected molecular weight — is what establishes what the material actually is, and a certificate reporting purity alone has not answered that question.

Two further limits are worth holding onto. Mass per vial is its own test: a vial can be 99% pure and still contain less material than the label claims, and every concentration figure on this page depends on the label amount being correct. And sterility, endotoxin, heavy metals and residual solvent screening are separately commissioned tests, usually priced individually — so a “third-party tested” badge asserts none of them unless the certificate names them. Check that the batch or lot number on the document matches the vial in front of you; an unmatched certificate describes someone else’s material.
